RAB GTPases in endosomal sorting and lipid transport at inter-organelle contact sites: implications in neurodevelopmental diseases and cancer

Membrane trafficking events are essential for cellular homeostasis and rely on the coordinated action of various regulatory proteins. Among these, RAB GTPases function as molecular switches that coordinate exchanges between intracellular compartments. Our lab is focused on understanding how RABs modulate crosstalk between organelles. We primarily investigate sorting events at early endosomes and have uncovered a novel role for RAB21 in regulating WASH complex activity. This function is critical for cargo sorting and recycling to the plasma membrane. Mechanistically, we identified a direct interaction between activated RAB21 and SWIP, a subunit of the WASH complex. Additionally, our proteomic studies on RAB GTPases revealed proximity between specific RABs and members of the lipid transfer protein family, ORP. These lipid transfer proteins are notably associated with inter-organelle membrane contact sites. To further characterize the protein composition at RAB-ORP contact sites, we developed a split-proteomic approach and identified a diverse array of proteins localized to these membrane contact regions. Ongoing studies aim to assess the functional impact of these proteins in membrane trafficking.
