” Transcription and evolution: the search for new regulatory elements in the genome “

Non-coding DNA accounts for over 98% of eukaryotic genomes, and contains a myriad of elements, some of whose role in regulating gene expression remains poorly understood. Characterizing these regulatory elements and understanding how they evolve are major challenges that need to be addressed. Indeed, several studies on animals and plants have demonstrated that most of the genetic variants associated with diseases or other phenotypes are located outside the coding protein genes, often in regulatory elements. The identification of relevant non-coding sequences and their mechanisms of action are often studied using two types of approach: genomic signatures under stress and evolutionary signatures. Our research group uses systems biology approaches that combine functional genomics, epigenomics and evolution to explore the functionality of noncoding DNA, particularly in response to stress. We use the genome of plants as a model for study, particularly Asian rice (Oryza sativa), a staple food for half the planet, whose production is impacted greatly by extreme weather conditions. In this presentation, I will discuss the characterization of enhancers, particularly those that produce transcripts as a sign of activity in a spatio-temporal context, as well as transposable elements that are sources of genes and regulatory elements. The approaches and technologies presented can be used in various research systems, from humans to plants.
